Abstract:

To increase the spectrum and sensitivity of analysing pharmacological active compounds, an optimised method was validated. Sample preparation is quicker, easier, rugged and efficient due to small changes of the EURL-method by Dendougui-Tadjine et.al [1]. A different extraction method with acetonitrile under sodium sulphate addition and replacing the solid phase extraction (SPE) step facilitates a big simplification. Instead of SPE a freeze-out step was installed. Additionally the extract is resolved in two ways. Therefor the extract is split in two, evaporated to dryness and resolved in two different solvent-mixtures. The two different extracts lead to additional clean up and optimal conditions by aligning the solvents to the LC eluents.

Validation was performed according to commission Implementing Regulation (EU) 2021/808 with muscle of fish, poultry and mammal (eight different species). Additionally an extension of method as regards to matrix was carried out for egg.

49 analytes have been validated - coccidiostates (ionophore und chemical coccidiostates), nitroimidazole (prohibited for food-producing animals) and selected pesticides (used against coccidia).

The method fulfils performance requirements of confirmatory methods according to IR (EU) 2021/808 for all 49 analytes in muscle and egg.
